BreastOncPx™ (Breast Cancer Prognosis Gene Expression Assay) 

US LABS now offers a 14-gene signature assay, BreastOncPx™ that provides prognostics information for lymph node-negative (N-), estrogen receptor-positive (ER+) breast cancer patients and is associated with risk of distant metastasis.  It helps identify higher-risk patients who might benefit from additional therapy.

Many breast cancer signatures have been published including the 70-gene MammaPrint® microarray-based signature, a 21-gene Oncotype Dx® RT-PCR signature, a wound-response classifier, an “invasiveness” gene signature, a two-gene HOXB13/IL17BR expression signature, and an intrinsic gene classifier that identifies subcategories of breast cancers such as basal, luminal, and HER-2.  Using a single data set, Fan et al reported that even though they contain different gene sets, the more complex signatures showed substantial agreement in outcome classification.

Specimen Requirements

Specimen:  Primary breast tumor: Send five precut unstained slides from paraffin block in sections and an H&E reference slide or formalin-fixed, paraffin-embedded (FFPE) tissue block containing ≥50% tumor. Zinc formalin fixative is detrimental to optimal RNA recovery. Please submit the regular buffered formalin specimen for this test.  

Volume:  Five unstained slides and one H&E stained slide at 10 μM or formalin-fixed, paraffin-embedded tissue block  

Minimum Volume:  2 mm x 2 mm tumor area  

Container:  Slides, blocks

Causes for Rejection:  Tumor block containing insufficient tumor tissue; tumor fixed in heavy metal fixative; broken or unstained slides
